DC1: Role of chromatin factors in establishing nuclear mechanical properties – UNITN

DC1 – (UNITN) will determine the functional connection between chromatin compartmentalization and nuclear mechanics in CPs. DC1 will develop MSC-based models of CPs by adopting CRISPR/Cas9 technology to insert truncating mutations in the causative genes of CPs. DC1 will characterize the obtained MSC-based models concerning chromatin organization, nuclear architecture, and mechanics. DC1 will also assess the responsiveness to mechanical cues stimuli controlling MSCs lineage commitment. Finally, DC1 will set up a high-content screening approach to identify drugs that may re-establish the proper chromatin organization and nuclear mechanical properties, thus designing new and effective drug treatment for treating CPs.
